Macromolecular drug delivery via transmucosal routes has been an interesting but challenging task for the researchers since the discovery of first therapeutic peptide decades ago i.e insulin in 1922. Selection of an appropriate carrier system is very critical aspect for safe, effective and convenient means of the therapy via transmucosal macromolecular delivery. Physiochemical complexities and biological properties of macromolecular bioactives demand an efficient carrier system that must address the issues such as molecular size, conformational stability, biological half-life, immunogenicity, dose requirements, complex feedback control mechanisms, susceptibility to break down in physical & biological environments; requirements for specialized transport mechanisms across biological membrane are duly considered.
